Post Transcriptomics

Transcriptomics
It is a study of transcriptome where as transcriptome is a complete set of RNA transcripts.
Transcriptome
It is a set of all RNA types which includes
1-m RNA
2- r RNA
3- t RNA
4- NON coding RNA etc
Relation of transcriptome and proteome
Transcriptome is a precursor for proteome. The proteome is expression of genes through the proteins, in other words proteome is the entire set of proteins expressed by genome.
Salinity
The salinity stress is observed at cellular level, organ level and whole plant level, and plant growth responds to salinity in two phases,
1- Osmotic phase
2- Ionic phase
1- Osmotic phase
Osmotic phase is that phase which reduced the growth of young leaves and makes them slower in growth.
2-ionic phase
Ionic phase is the phase of removal of mature leaves.
Plants have adaptations to salinity and these of three type
1- Osmotic stress tolerance
2- Na+ and Cl- exclusion
3- Tolerance of tissue accumulated to Na+ and Cl- ions
Salinity transcriptomics
In the short, It is a study of genes and their expression under the salinity stress conditions.
Example of salinity transcriptomics
Wheat crop is one of the most important food crop in the world, the transcriptomics analysis of that crop which promise us to describe the expression dynamics of genes these genes may control many agriculturally important traits.
Recent work done in salinity transcriptomics on different plants
Cultivated tomato is the most important vegetative crop, which is moderately sensitive to salinity To improve the salt tolerance in tomato cultivars, wild halophytic tomato has used as ideal gene donor because wild halophytic tomato genotype , Solanum pimpinellifolium is significantly more salt tolerant as compared to cultivar.
To observe the transcriptomics, the affmetrix tomato genome array containing 9200 probs sets was used to compare genome activity with pl365967 and moneymaker. The results of treatment showed that
pl365967 expressed a few responsive genes as compared to moneymaker and salt sensitivity was observed to be more active in pl365967 than in money maker .Less accumulation of Na+ was noted in
shoots and roots of pl367967 due to salt tolerant genes encoding lactoylglutathione lyase.
H. CHELAIFA etal stated that invasive species are much important and best model to investigate the
Evolutionary processes associated with ecological success. We can judge ecological success by
Comparisons with the closely related species and here have to describe the transcriptome evolution
Following divergence b/w two closely related salts –marsh species Spartina alterriflora is invasive species
and it is native to the East-American Atlantic coast and now it is introduced into several continents .
We investigate through cross species hybridization micro-arrays using rice oligo micro-arrays to describe
and to make comparison from 10 nuclear genes (2256 bp) revealed that nucleotide divergence b/w
spartinar and rice ranges from 8% to 14% and 44.5%(9353) genes is hybridized on array hybridized with
both species.
